Some authentication modules have the ability to individually specify a non-default location for an account's maildir. A home directory must still be defined, but the account's mailbox can still be specified. Note that the account's mailbox must be separately set for each account. There is no single button to push that will place all maildirs in /var/mail.
I'm confident that if Postfix supports maildirs, it should have a straightforward setting that places all maildirs in $HOME/Maildir. Maildirs predate Postfix's support, so whenever Postfix added support for maildirs, it must've been able to do that with a single setting that puts all maildirs in $HOME/Maildir.
