I'm looking for a way to convince maildrop to create a Maildir if it doesn't exist. For example, I'm adding a new entry into my mysql authentication database and I want that maildrop will create the maildir on the first delivery. Is that possible? If so, is it possible to use a template directory?

No, it's not possible with maildrop.
Try having it done through the system, /etc/skel for example, or trigger it off mysql somehow...
