MAILDIRPATH=./Maildir

MAILDIRPATH=~/Maildir

The syntax isn't "weird", it's shell script. If you use "~/Maildir", the value will expand to "/root/Maildir" when the service starts, and every user will try to open that path.

MAILDIRPATH is "Maildir" by default, which is the path, relative to the users' directories.
