AFAIK, only global filtering does that. However, it cannot alter messages.
If the server you forward to can be found via MX you need no other setup.

If you want to deliver locally without defining local mailboxes, you
can install a .courier-default in aliasdir. If you do that, then you
_must_ set your maildropfilter file in order to check that the remote
mailbox exists, e.g. http://www.courier-mta.org/localmailfilter.html
