Your limiting factor is going to be bandwidth. There's nothing really specific to tweak, up front. Just being with the default config, and go from there. Your tweakable knobs are going to be in the modul.esmtp configuration file. See the slightly obsolete documentation in http://www.courier- mta.org/modules.html, namely the "config" section.
One other thing you may or may not need is a second machine, where messages that cannot be delivered in a reasonable time will get forwarded to, to sit and stew for a while, and not clog up your main mail server. Use the backuprelay config setting, documented in the courier man page, for that.
