Yes. ZODB Directory is designed as a stopgap solution for small directories for people who don't want the hastle of setting up an SQL database. But for 50000 people, you should really use SQL (or LDAP). In theory ZODB Directory could be made fast, but a dedicated indexing catalog for it would have to be written and used so that searches are fast.
