I was able to boot Knoppix 3.8.2 from a CD and then continue from an NTFS partition. The following was done under Windows XP Pro:

1) download Knoppix 3.8.2 ISO image, put it in c:\, and name it knx.iso
2) place a linux swap file in c:\ and name in knoppix.swp
3) place a Knoppix 3.8.2 persistent home image in c:\ and name in knoppix.img
4) reboot with the Knoppix 3.8.2 CD in the CD-ROM
5) at the boot prompt ...
boot: knoppix bootfrom=/dev/hda1/knx.iso
6) select "OK" when Knoppix asks about using the persistent home image

Advantages of this method over using the CD alone or using the CD+USB stick:

- faster than booting from CD alone
- acts like a read/write Linux system
- no need to repartition NTFS drive
- multiple *.img files for different uses. Copy desired image to knoppix.img while in Windows
- allows ejecting of CD. Useful if you have only one CD/DVD/combo/ super drive.
- uses the swapfile so that more RAM-intensive applications can be run.
- no need for the USB stick (if you want to copy the image, copy from within Windows)
- if you turn on the Hidden flag under Windows, no one will see the files from within Windows

Disadvantages:

- you need to boot to Windows between successive Knoppix boots so that Windows can run chkdsk (dunno why, yet.)
- if Knoppix crashes, the persistent image file could get corrupted.

Wishlist:

- a Windows utility on the Knoppix CD for creating the knx.iso file
- a Windows utility on the Knoppix CD for creating the knoppix.swp file
- a Windows utility on the Knoppix CD for creating the knoppix.img file
- a kernel boot alias that boots with the above options
- coLinux booting from a Knoppix image.
