long exposure will make a difference (for canons that the in-camera
black frame subtraction, which also affects raw). it will definitely
be a good idea to do both measurements (that on and off) and maybe
even with a couple of different exposure times. this kind of noise
(gaussian part) should mainly be visible for long exposures, which is
also where the black fram subtraction kicks in most. i don't expect
the poissonian part to change much though.
