I'd be careful with exposure lengths that long, you're pretty likely to get
smudging/star trails due to the earth's rotation. But it really depends on
your focal length. The "Rule of 500" gives a decent guideline:

Definitely go with manual exposure, and manual focus as well. The comet is
pretty bright, as long as you have a large aperture you probably won't need
very long exposures.
