The reason why I didn't mentioned any MUA is because all used MUA here 
showed the problem. (kmail, evolution, thunderbird, mutt, pine)

Also it was reproduceable using "telnet server 110":

You see, No eMail is availible for user elke.

Now the same thing with "default_mail_env = mbox:/var/mail/%u" setting:

server:root# /etc/init.d/dovecot stop
Stopping mail server: dovecot .
server:root# vi /etc/dovecot/dovecot.conf
server:root# /etc/init.d/dovecot start
Starting mail server: dovecot.
server:root# telnet server 110

Connected to server.s.automatix.de.
Escape character is '^]'.
+OK dovecot ready.
user elke
+OK
pass ******
+OK Logged in.
stat
+OK 19 150212

So this error must occour with all known POP3 Clients.
