I have been suffering from the same problem for a few days now. In my case it was caused by an (presumably) invalid backend for Debian-Security (maybe due to updates on the mirror). The mirror I used was http://ftp2.de.debian.org/debian-security.
This caused the daily cronjob for apt-cacher-ng to malfunction. Using the "Start expiration" button in the web interface didn't work either. By switching the backend to another mirror it solved the problem for me. Nevertheless, this could be a bug in apt-cacher-ng.
