Tried to upgrade OTRS from 2.4.9 to 3.0.8 (unstable) without dependencies to
unstable.
After updating OTRS said JSON.pm missing in @INC. But JSON.pm was there, only
json.pl was missing in /cgi-bin/ I couldn't solve the problem. Thus I
downgraded. That worked fine.
I suspect that /usr/share/otrs/Kernel/System/JSON.pm is not the JSON.pm
from the error message. Is the package "libjson-perl" installed? if not
-> install it. :)
If this package fixes your problem, i'll add it to the dependencies.

PS: otrs 3.0.x needs the libcss-minifier-perl &
libjavascript-minifier-perl package too - both not available in squeeze.
