They don't run automatically during package build for either pbuilder or sbuild afaik, although that would be really nice. Ubuntu's buildd's do run them during the publishing phase, and you can run them locally, which isn't a bad idea. Here's Ubuntu's documentation on autopkgtest, which seems the best so far.
