I'd heard through MS that Vista wasn't going to support 16-bit apps. It seemed to be a factual type statement along the lines of "no it won't because none of the code to support 16-bit is in there". This was in the last month. That being said though I've not even installed Vista anywhere so can't and haven't tested this.
