This requires imagemagick, though, which isn't installed by default.
However, why not use GNOME's own thumbnailer? It also converts
transparency into white:

$ unzip -p /usr/share/example-content/oo-maxwell.odt Thumbnails/thumbnail.png > 
/tmp/thumbnail.png
$ gnome-video-thumbnailer /tmp/thumbnail.png /tmp/nautilus-thumbnail.png

/tmp/nautilus-thumbnail.png looks just fine to me.
