Sebastien,
Per your request, I have downloaded the "karmic-desktop-amd64.iso" image 
from "http://cdimage.ubuntu.com/releases/karmic/alpha-2/";, burned it to 
CD and used it to boot my computer, the same machine that the problem 
was reported against. After booting, to enable DVD playback, the 
"***ubuntu-restricted-extras package 
<apt:ubuntu-restricted-extras?section=universe?section=multiverse>" was 
installed from location 
https://help.ubuntu.com/community/RestrictedFormats.*

Sound operation was verified good.
A movie DVD was inserted into the second optical drive.
A program selection popup panel appeared, "Movie Player" was selected.
Video was displayed but there was no accompanying sound.
Movie Player was shut down.
DVD contents were opened in a file browser.
After navigating to the video folder a .vob file was opened with Movie 
Player.
Video and sound played normally.

In other words, the problem still exists in Ubuntu 9.10.
