I haven't used the Application scope since finding out about the Request
scope. However, the Application.cfm file on one site has swollen to the
point where there's about 20 structures containing site wide data in the
Request scope, some containing multiple structures or arrays.

Is there a rule of thumb which says "once you get to point x, it's going
to be more efficient to store them as Application variables". Now we
don't need to lock reads and writes, should I shift them back to the
Application scope?

I'm redoing it at the moment, now's the time to change it if it would be
better. What do you reckon?
