Middle tier would be any other third party product / tool such as Java
itself (most probably JRun or any other J2EE server).

"Understanding of how Coldfusion interacts with Middle Tier is required."

What do they mean here?

After some preliminary research, it would seem that CF, as an application
server, *is* the middle tier: the middleman between the client and the data.

Since I am primarily self-taught on CF, it seems this kind of terminology is
one of those things that I haven't been exposed to yet.
