Route targets are used in the typical manner of MPLS networks to define network reachability, but you only have to specify them explicitly when you interoperate with BGP routers that are not under Contrail's control. It's not a matter of physical vs virtual. Any non-Contrail router that you're using, whether physical or virtual would need to be configured with matching route targets in order to exchange routes with Contrail. Contrail also uses route targets for vRouter to vRouter networking but it manages them internally so you don't have to explicitly allocate and configure them.
Generally you'll want at least one gateway between your Contrail networks and all the rest of the world (whether that's the Internet or one or more private networks whether WAN or LAN), so you'll want to coordinate what route target(s) to use between Contrail and that gateway.
