Yes. This is implemented for all subclasses of the HTTPRequest layer -- if the "URL" parameter is an Array (as described as a possibility on http://dev.openlayers.org/apidocs/files/OpenLayers/Layer/HTTPRequest-js.html#OpenLayers.Layer.HTTPRequest.OpenLayers.Layer.HTTPRequest), then a URL will be selected based on a hash of the tile parameters. The multiserver.html file demonstrates this.
