Yes, everything in the Release Notes! :)

As a general rule, the Release Notes are here to explain the important changes 
and thus what needs to be tested.

Now for some of them you may not know how to test and you could ask specific 
questions to the devs on IRC or the list.

If you look at the RN for 5.0M2 for example 
(http://www.xwiki.org/xwiki/bin/view/ReleaseNotes/ReleaseNotesXWiki50M2) you'll 
see:
* Automatic paste: needs to be tested on all supported browsers
* Virtual mode on by default: try creating new wikis, new workspaces, etc. XEM 
should be installable without restart now too.
* Try accessing an unknown subwiki and verify the message displayed
* Be careful about authors/creators information displayed as changes have been 
done related to that
* Sergiu has made a DB schema change with a migration so upgrades from older 
version to 5.0M2 on all supported DBs should be tested 
* Translations for workspaces
