Yes, you need to compile your application either for gtk+-x11 or for
gtk+-directfb. This is not a matter of exchanging the library, you
have to recompile the applications that use it.

directfb-engine is just a GTK+ theme engine similar (and even
file-format compatible with the pixbuf-engine). It's not required in
any way.

I am not sure if you can get any GNOME applications compiled on
gtk+-directfb. As far as I know the basic GNOME libraries depend on
X11. It might be possible to hack around this dependency by removing
some functionality but you can not expect to be able to run GNOME on
DirectFB without any changes (unless you want to use XDirectFB).
