Even with locking, NFS will be problematic. You have machineA and machineB mounting a NFS share with your mail store, and let's say you're using Dovecot because I know it stores index files in the Maildir directory. If you have locking enabled and you try to simultaneously run mail programs on both machineA and machineB, one will hang waiting for the other to release the locks on these index files. If you don't have locking enabled then the two concurrent instances will trash each other.
