OT: A left hand perspective. 

"As soon as left mouse button is pressed down, the cursor turns to a
closed hand,"

As a lefty I use the right mouse button. This concept is confusing at
times but I am used to it as I have had to adapt to this right handed
world.

As for the drag and drop versus buttons or double clicking. I like
being able to double click a selection and have it populate in a
different text box. This takes the least amount of time and energy
because it doesn't require mouse movement or moving to a button and
clicking. As long as there isn't the annoying page refresh after
each item is moved over double clicking is my choice.
For keyboard accessibility one can tab into the field, scroll with
arrow keys to select text and "enter" moves the selection. Or left
and right arrow moves the selection from one to the other and back
again.
