Personally, I'd do this with two disks, a 2.5" for the work music and
3.5" for the flac library backup.  For a couple of reasons.

A 2.5" notebook drive is more durable and designed for that sort
portable use.  It's smaller and easier to carry around.  And you can
most likely run it without an external power adapter, which makes it a
lot easier/faster to attach and detach from your PCs.

The flac library backups don't have to be done all that often unless
you're involved in a flurry of CD ripping.  Run a backup once a week,
or once a month and then store that drive offsite.  You'll only need a
drive the same size as your internal flac music drive instead of one
large enough to store both the flac and mp3 mirror library.  So when
you go to upgrade to (say) a 500GB disk for the flac library, you can
just buy two of them and have your backups covered.
