You should use Task Manager or download Process Explorer to check for a process called Slim.exe. If it is not running then Slimserver is not running regardless of what SlimTray say.
In 6.5.1 - depending on how Slimserver is set up and started SlimTray is unreliable when reporting whether Slimserver is running or not. You should upgrade to 6.5.2 to fix this problem.
