The iTunes store location you use is based on the credit card billing
address location.  If you're using a UK billing address, you should 
use the UK store.  That might be set up automatically when you 
set up your account.  If you try to buy items in the U.S. store, you'll
get a message like "Your account is only valid for the U.K. iTunes
Store.  Clicking OK will take you to this store."

After the initial "review" of account details where you input your
credit card details, you don't need to enter your credit card again.
You just need your login name and password to validate purchases.

If you're in the UK store all the prices will show up listed as UK pounds.
What I'd suggest is that you "practice" purchasing the Free UK iTunes
song after you set up your account. This is like doing a purchase, but
for 0.00 pounds (or dollars), and will take you through the process
without the possibility of getting charged for a mistake.  In general,
you can find the page for this by googling "macrumors free iTunes 2007"
which takes you to the U.S. entry that lists all the free iTunes songs
posted so far for 2007 (a pretty long list).  The post lists International
lists in this thread.  I'll give the links for the U.K., Canadian, and 
Australian stores (but it's easier to google for the information):

These lists get updated once a week, usually after Tuesday night, and
generally only the most recent week's listing will be free. (Sometimes there
are codes like [DD] = Discvery Download or [SOT] = Single of the Week,
and now [LTSOW] = Latino Single of the Week).

When you purchase a song, by using the button to get the song in the
column where the prices are given, you'll be prompted to either create an 
account, or to log into your account with username and password.
If you're already logged into your account, and some time has passed,
iTunes will ask you to enter your password. You don't need to do this
again for the next few purchases unless a long time has passed.

Once you've purchased a song, you can check that the download has
started by tabbing to the Sources Outline and typing "Dow" to move
to "Downloads"  (which appears when a download starts) and hear
"Downloads 1 item" (or however many are in progress at the time).

In the case of free songs in the middle of an album list, the link
will take you to the free selection.  So you should be in the Songs
Outline at the right song; just start interacting, and then VO-keys right
arrow over to the Price column. (You can check this with 
VO-keys shift-C).  If I have Mouse cursor tracking VoiceOver cursor
on, all I have to do is use VO-keys shift space to select the song
for purchase on my 12" G4 Powerbook.  Maybe wider screen
displays stretch out columns so that the mouse click on the center
of the column doesn't fall on the button to purchase?

Using the Search Text menu works pretty well, because it gives
you results in order of popularity. However, if you want to search
with more control, you can try using the Advanced Search link
for music from your browser:

Now if you move from the Search Text Field (option-command-F) and tab
to get to the Songs Outline, after getting "iTunes Store" your next four tabs
will put you into four unlabelled fields:  Artist, Composer, Song,
and Album.  Enter any selections you want  in any of these fields and
carriage return, but remember to clear each field of previous search
terms if you go back to these.This works for things like finding out which 
artist versions
of a song are available or getting a specific album, when the album name
is similar to a song name.  Now when you do your next tab to the Songs 
Outline your results will reflect searches where the terms are limited
to specific categories.  I'll mention that users can also use column sorting
to order search results -- different Artists who have recorded a particular
song, versions classified in a different genre, listings by Album, etc. but
you probably don't want to experiment with this when you're just beginning
to use the iTunes store.  It's better to try this out on your Music Library.

Sometimes the iTunes store is busy, and you won't get a response or
the connection will fail.  This usually happens just before they're adding
new features or doing major updates before an event. In those cases,
using the Advanced search browser links will also fail to connect.

I don't usually use the Advanced search music options at all.  This kind
of link is more useful for items like Podcasts.  I've also found that some
times when the Music part of the store is too busy to connect, I can still
get in with the Advanced Search links for podcasts or audiobooks, because
these parts of the stores are not busy.

Oh, and I don't know of a general accessible way of changing the store location.
Going to a song that is only offered in the UK store or the Australian store, 
etc.
by using the free iTunes offering will work,  You get a message that your
request could not be completed because the item is not in the (US/UK/etc.
store) but it is in the (other) store, and do you want to go there? if you want 
to 
browse in other stores this is one way to do it, but you won't be able to 
purchase 
from iTunes stores for other countries.  In fact, all your searches
will be run for that store location.  Trying to purchase a song from another
iTunes store will bring up a message like "Your account is only valid for the
xxx iTunes store. Do you want to be taken there?"
