Barney Boisvert did a great preso at cf-Objective() about running on EC2. Their company migrated their entire production environment from Rackspace's cloud to EC2 in a very methodical month long process that he went into some amount of detail about. His slidedeck is here:

So you really get a whole lot of bang for your buck. He discussed some issues around performance and deploying to multiple zones to avoid Amazon EC2 downtime.
