It doesn't work that way with Django. You run each Django project on a different port, and your Web server (nginx, apache, whatever) forwards incoming traffic to the correct port.
Your Python code (all Django is Python) should never be accessible via your Web server. Your Django app will run as a long-running process via gunicorn or a wsgi or an alternative.
