The amen break sounds like it does because it's a sample off a 1969 record by The Winstons called 'Amen, Brother'.  To recreate that sound you'd have to use a similar set up, i.e. Drum Kit, Drummer, Studio Equipment, Mics, Processing etc.  Now I'm assuming that's not likely, so therefore it'd be easier to sample the original, if that's the sound that your after, or else to create some different breaks that will have there own sound.

Lyn Collins :- 'Think (About It)'
Bobby Byrd :- 'Hotpants'
Michael Viner's Incredible Bongo Band :- 'Apache'

Dennis Coffey :- 'Scorpio'
Kool And The Gang :- 'N.T.'
Skull Snaps :- 'It's A New Day'
 
There are loads more but this should do for starters,
