Go to the web site's properties in the IIS manager and look at the
directory security tab.

Additionally, you may need to give the user IIS is using write access
to the temp directory: Temporary ASP.NET Files inside the
Microsoft.Net framework folder for the framework you are using.

You can also get more information to troubleshoot your issues by
looking in Windows Event Viewer.
