One way would be to use the 'bin/dspace user' command:
https://wiki.duraspace.org/display/DSDOC6x/Managing+User+Accounts

If you have a lot of users to define, it may go faster to start bin/dspace
once and feed it a file of commands:
https://wiki.duraspace.org/display/DSDOC6x/Executing+streams+of+commands

See also https://wiki.duraspace.org/display/DSDOC6x/Command+Line+Operations
for general information about the 'bin/dspace' tool, including a nice list
of links to details for each command.
