No, you shouldn't have to. You will find that the "ant update" build backs up a number of the subdirectories and creates new directories, it does so for "bin", "lib", "webapps". You will want to run bin/index- init after your upgrade because that will build the browse and search indexes critical for DSpace to operate properly.
