Generally you shouldn't need to set a classpath variable. For the web applications (xmlui / jspui / sword etc) this is all handled by Tomcat by the way in which the web apps store classes in particular pre-defined places. For command-line applications (e.g. importing, filter-media etc), this is handled by the 'dspace' script (1.6+) or the 'dsrun' script (1.5.2 or earlier).
