Use the File/Databases/Labels and select one of the two custom labels that you can customize. I use one for my labels and one for printing directly to qsl cards. Use a blank piece of paper to check how the print layout is working until you get everything right.

How do I alter the layout? I don't see anything in the manual about the ability to actually rearrange the entire label layout process, i.e. a label layout or configuration tool.
