Is it a CT-17 interface or something else? If it's an after-market inteface (like K1NU or others), you may need to set RTS and/or DTR high in the radio setup in order to make the cable work.
Also make sure that the radio hex address in the program matches the one in the radio, the baud rates match (i.e. don't use AUTO on the radio), and that that you are running N81 (no parity, 8 data bits, 1 stop bit).
