Yes, you have to add it on every ltsp-server.
For more information on the syntax of group.conf read man group.conf -
in short, the meaning of
 *;*;*;Al0000-2400;plugdev,scanner,audio,fuse
is:
for all services (*), on all terminals (*), all users(*) on all days
(Al) 24 hours a day (0000-2400) should get the additional groups of
plugdev, scanner, audio and fuse. If you want to add plugdev group only
to a specific group (say, teacher), than you'll have to either list the
users explicitly or use netgroups. Unfortunately, you'll have to ask
somebody else about netgroups, I don't know where they differ from
normal groups.
