This is also happening to me, and has happended with all the Endian versions 
i've tried. 
For what i saw when freshclam finish to update the antivirus datatabase it stop 
the clamd daemon and then restart it. 
When clamd is down, amavisd fail to connect to it and then the die.

If you look the logs you can see that clamd stop at 09:20:22 and amavisd gives 
connection refused at 09:20:28, just 4 seconds later it can't connect to clamd. 

what i did today was to change the update to daily, instead of hourly, and edit 
the cron file (/usr/lib/clamav/freshclam.cron):

I did not run into this problem in 2.2.beta1. I see that this seems to be a 
problem in 2.2beta3. My work around at the moment was to disable smtp 
antivirus scanning and then all emails went through pretty quickly.
