Dave,

Yes, those locking screws should always be screwed in. Things may work without it in a temporary setting (for a quick test for instance), but for a permanent station setup, screw in those jackscrews. If the equipment is moved or there is anything that might "tug" on the cable or vibrate it, the jackscrews will keep it from being disconnected.
