Yes, you can achieve nice multiflash- setup alike shots by combining multiple
exposures with flash.  One way to achieve balanced lighting is to modify the ISO
to compensate for the multiple exposures in the same way you would for a
multiple exposure without flash. In that way you affect flash and ambient light
at the same time and by the same amount.  IF you want to simulate effects like
main flash and fill-in with some kind of ratios, then you have to apply flash
exposure compensation on top of it.

A simple example: You want to use two "lights" with ISO100 film. Then you set
multiple exposure to 2 and ISO200. If you want the lights to be balanced, shot
each of the two times by placing the flash and reflector pointing to your
desired side of the subject.
If you like main light and fill-in, dial +1/2 stop FEC before shoting the side
that should be fully lighted and -1/2 FEC for the fill-in light. The total
difference is 1 stop or a 1:2 ratio.

That's the idea. Now play with it to achieve what you have in mind.

Note that this method is also effective to simulate strobe effects with
flashguns without this feature.
