I might also add that performing mailbox-level backups rarely meets expectations and more often cause more problems than they are worth. If you would like more detailed information on the rant against brick-level mailboxes, I'll post the link.

This is very likely why the Mailbox level backup is failing. Mailbox-level backups (aka Brick-level backups) use a MAPI profile to log in to each mailbox, so the account you are running the backup under must have permissions to all mailboxes. Even if you have added the account to the Exchange Domain Servers Group, it still may be denied access because Deny will take precedence over Allow. This is normally why it is recommended to run services such as Backup software using their own dedicated service account.
