Are you using cached mode in Outlook? If not, do so.

Often, latency experienced by users is a consequence of Outlook needing to
render a view.  

Does a 5MB file stored on a file server open much more quickly than a 5MB
attachment?

Sometimes when someone clicks on an email to open it, there is some latency.
The director said that when he emails an attachment >5 meg, he says it'll
hang (but this is not an everyday occurrence).

We're running Exchange 2000 on a W2K server.  My director pointed me to
PerfectDisk by Raxco as a possible tool to "speed up" Exchange response.  I
was wondering if anyone is using this product and can let me know if it's
worthwhile to look into it.  
As I noted, E2K STD with program files on C, logs on D and mailbox store on
E (attached to SAN drive) and about 150 mailboxes..  I don't think defrag is
an issue as much as we're running anti-virus and anti-spam on the same
server.
