It is scheduled automatically, although you can change this by going to the
properties of your server in the admin program. This is online maintenance
so the services do not stop.

I forgot to add that I would stop the Exchange services before doing any
defragging.
This applies to file system defragmentation, by the way. Exchange
defragmentation happens during the IS Maintenenace period. I don't think
that's what you're asking about, though.
