Exchange 2000 does not apparently need the log on locally right for OWA to work. However, you really should look at the NTFS permissions on the /exchweb virtual directory. I experienced the same symptoms when, in an effort to tighten security, I removed the Everyone has Full Control NTFS right for this drive.
Did someone run IISLockDown on your web server over the weekend? You might try backing that out by running it again.
