For 40 users, I would seriously consider using exmerge to pst for mailboxes. Use Outlook to export public folders to .pst.
As you probably know, there is no direct upgrade path from 5.0. It requires 5.5 sp3 minimum. I see you are looking at moving to 5.5 first in your path, but now your are talking about 2 mailbox moves. If you are prepared for that, your plan is feasible. Do not underestimate the Active Directory requirements and read up on the Active Directory Connector before installing Exchange2000. I would then promote this machine to PDC. After the data has been moved, I figure I can upgrade (to windows 2000 server) the existing PDC and install exchange 2000 in the same site as the exchange 5.5 machine. once this is done I figure I can move everything to the exchange 2000 machine and drop the 5.5 machine from the site (once all the data has replicated/moved) Does this sound feasible?
