This doesn't have to be done on Exchange Server at all. If they don't use NAT, then all you need to do is route their MX servers on your Router/Firewall/VPN end point to route the traffic through the tunnel.
If they do use NAT, they you probably have to set up host files on your exchnage server to indicate the appropriate mx server addresses. Either way, it's probably best to deny the access to those addresses from your outgoing border routers to ensure that no mail went through that way.
