It will be difficult, but possible. By creating distribution lists (I assume you're talking about Exchange 5.5) and using the Delivery Restrictions tab of each recipient's mailbox and in the Internet Mail Service's address spaces, you can restrict who can send to whom and to the Internet. But it will be a lot of work. There's no way I know of to do this on the sender mailbox objects.
