Kerning is the spacing between each character. When you have kerning turned on it will adjust the spacing between character so that they look more uniform. The amount of space needed before and after the letter 'T' is much different than that needed for 'G'. This is because the 'G' is more bloated at the sides where 'T' is more narrow at the side. So, you can really but-up a letter before and after a 'T' where as you will need more space before and after a 'G'.
From dictionary.com The process of reducing the spacing between certain pairs of letters to improve their appearance. When a font<http://dictionary.reference.com/search?q=font> is created, each character<http://dictionary.reference.com/search?q=character>is given a width that includes some space around it so that the letters don't run into each other when displayed or printed. This can be thought of as an invisible box around each character. Some pairs of characters such as A and V, look better if the boxes overlap slightly, bringing the characters closer together (but still not touching). Charles P.
