I think the main pre-requisite should be some knowledge of the concept OOP.
It's a well written book, but OOP can be hard to grasp at times.

If you worked with classes etc using AS 2.0 you should be fine with this
book.
It explains some of the biggest changes in the first few pages off the book.

The book mentioned in one the replies "head first: design patterns" helped
me
out with understanding what design patterns are etc.
I know nothing about java, but that wasn't a problem at any given time.

I would suggest:

1/ read the head first: design patterns book (or follow a basic Java course,
it's really helps when working with AS3)
2/ buy the actionscript 3: design patterns book by Joey Lott and David
