Make sure you have cvsup

$sudo portinstall cvsup-without-gui

check your supfile

$cat /usr/local/etc/cvsup/supfile
#
*default tag=.
*default host=cvsup5.freebsd.org
*default prefix=/usr
*default base=/usr/local/etc/cvsup
*default release=cvs delete use-rel-suffix

src-all tag=RELENG_6
ports-all
#

update your source tree

$sudo eval `which cvsup` /usr/loca/etc/cvsup/supfile

also, make sure you are in the right directory with the "Makefile" for
building the iso.
